中文摘要: WRKY转录因子是植物响应病原菌胁迫最重要的转录因子之一,参与抗病反应及信号传导通路的调控。项目组前期已利用cDNA-AFLP技术获得了一个不结球白菜响应霜霉病胁迫的BcWRKY1转录因子,并发现BcWRKY1属于不结球白菜对霜霉病菌早期应答的基因,但其结合特性、抗病功能及调控机理尚不明确。本项目拟采用转基因技术对高感材料进行BcWRKY1过量表达和对高抗材料进行RNAi抑制表达,对BcWRKY1抗病功能进行分析,并利用荧光定量PCR技术分析BcWRKY1转录因子对PR等防御反应基因表达的影响;同时,利用酵母双杂交等技术筛选BcWRKY1互作蛋白,并利用BiFC等技术对其互作进行验证,探讨BcWRKY1转录因子在不结球白菜抗霜霉病反应中的抗病功能及调控机理。通过本研究,不仅有利于阐明不结球白菜抗霜霉病的分子机制,还有利于推动不结球白菜抗霜霉病分子育种进程,具有重要的科学意义和应用前景。

英文摘要: The WRKY transcription factors are most important family transcription factors of plant in response to pathogen, with roles in disease resistance and signal transduction pathways. We obtained the BcWRKY1 transcription factor gene using the cDNA-AFLP in non-heading Chinese cabbage, which is responsive to downy mildew. We found that BcWRKY1 is an early responsive gene to downy mildew, but its DNA binding properties, functions and regulation mechanism in disease resistance are not clear. The project will analyze the function of BcWRKY1 in downy mildew resistance by using over-expression transgenic plants and RNAi silencing plant, and identify proteins which interact with the WRKY transcription factor by using the yeast two-hybrid system and BiFC technology. The results will not only help elucidate the resistance mechanism to downy mildew in non-heading Chinese cabbage, but also help promote the process of molecular breeding in antidisease.
