中文摘要: 楸树是梓树属中重要的栽培树种之一,其生长速度快和材质较好。随着经济的发展,我国对楸树的需求越来越大,但是楸树扦插生根困难,嫁接育苗是目前主要的育苗方法。但是该方法存在育苗成本高,病虫害严重等问题,限制了楸树的大面积推广。因此揭示楸树嫩枝扦插生根的分子机制并培育易扦插生根的楸树品种是目前急需解决的问题。由于楸树的研究基础较差,对其扦插生根的分子机制的研究还未见报道。本研究以扦插生根率高的楸树品种豫楸1号为研究对象,利用RNA-seq测序对不定根发育的4个时期的转录组进行BLAST、GO及KEGG等分析,发掘调控不定根发育的主要的代谢途径和重要的基因。同时设计EST-SSR引物结合关联作图,发现与不定根发育相关的贡献率大的QTL。本研究目标是发现楸树嫩枝扦插生根的基因调控网络,发掘与扦插生根率相关的贡献率大的QTL,从而为利用基因工程育种手段改良楸树提供科学依据,具有重要的科学意义和应用价值。

英文摘要: Catalpa bungei is one of the important cultivated species in the Catatpa scop.Catalpa bungei is characterized as fast growing and excellent wood qualities but the rate of adventitous root of the softwood cutting is low.With the development of economy in China,the requirement for Catalpa bungei was fast increased.Now, most seedlings of Catalpa bungei were generated by grafting,but high costing and serious disease were the major disadvantages. Therefore,it is very important for Catalpa bungei breeding to illustrate the molecular mechanism of adventitous root of softwood cutting at present. Up to now, there was not report on transcriptome analysis of adventitous root of softwood cutting of Catapa bungei because of poor research foundation.In this study,transcriptome of adventitous root during the four stages of softwood cutting in Catalpa bungei was researched using RNA-sequencing.The major objective of transcriptome analysis was to find primary the signal pathways and important genes through bioinformatics tools and methods (BLAST, GO and KEGG).The softwood cutting rate, number of adventitious root and length of the longest adventitous root was investigated in this study.
