中文摘要: TGA转录因子通过上调PR基因的表达增强植物的抗病性。项目组通过酵母双杂交筛选从苹果中分离了1个TGA转录因子MdTGA2.1,并构建了MdTGA2.1的过量和反义表达载体,而且已获得了转MdTGA2.1-GFP基因的苹果组培苗。在此基础上,本项目拟转化苹果和拟南芥突变体,通过转基因苹果的抗病性鉴定以及拟南芥突变体的功能互补试验,鉴定MdTGA2.1的抗病功能;利用转MdTGA2.1-GFP的苹果材料,通过ChIP-Seq技术在全基因组范围内寻找MdTGA2.1调控的PR基因,并利用ChIP-PCR、EMSA和qRT-PCR等技术证明MdTGA2.1对PR的调控作用及PR基因对病菌侵染的响应。通过本项目的实施,不仅能阐明MdTGA2.1的抗病功能,而且获得的PR基因可以作为防御激活的标记基因,为苹果中其他抗病基因的功能鉴定及抗病机理研究奠定基础,并为苹果抗病育种提供理论依据。

英文摘要: TGA transcription factors enhance disease resistance through positively regulating PR gene expression in plants. In the preliminary study, MdTGA2.1, a TGA transcription factor in apple, has been isolated by yeast two-hybrid screening,and over-expression and antisense vectors have been constructed. In addition, the MdTGA2.1-GFP transgenic apple plantlets have been obtained. On this basis, we plan to stablely introduce vectors into apple and arabidopsis mutant,and then verify the function of MdTGA2.1 through disease resistance identification of transgenic apple plants and the functional complementation experiments of arabidopsis mutant. To identify candidate PR genes regulated directly by MdTGA2.1, we will search in whole-genome of MdTGA2.1-GFP transgenic apple with chromatin immunoprecipitation sequencing(ChIP-Seq). Then ChIP-PCR, electrophoretic mobility shift assay (EMSA) and real-time quantitative RT-PCR will be performed to verify regulation MdTGA2.1 to PR genes and response of PR genes to infection.Through the implementation of this project, the function of MdTGA2.1 in disease resistance will be characterized, Furthermore, the PR genes obtained in this study,as marker genes for defense activation, will play important roles in identifying the resistance function and mechanism of other resistant genes,and prov
