中文摘要: 随着高通量测序技术的发展,越来越多的研究认识到细胞内部转录产物十分复杂。我们早期的研究表明在细胞内,既有大量未知的非编码转录本存在,同时编码RNA 的转录形式也是多样的,但是只有少部分的转录本会用于蛋白的编码和转录的调控,对于所有的转录产物最终的作用区域和意义尚不了解。我们以小鼠胚胎干细胞为材料,分离、收集在细胞核、细胞质以及多聚核糖体中的RNA,应用高通量RNA-seq 技术深度检测各自部分的转录组组成,结合公共数据库中胚胎干细胞的表观修饰数据以及早期关于细胞整体的转录组数据,研究转录本在细胞内部的分布及意义,区分不同类型转录本在基因组上的特征,研究在复杂转录背景下真实有效的转录本的特征,为构建模拟细胞调控网络提供基础。

英文摘要: The development of high-throughput sequencing technology has greatly pushed the concepts that there is a quite complicated transcriptome in cells. Our early-years research showed the existence of a large number of unknown non-coding transcripts and the variety of forms of the coding RNA. Only small fraction RNA will be used for the protein coding and transcription regulation. but regional distribution and significance of all the transcripts are unknown. We separate and collected the nucleus, cytoplasm and polyribosome from mouse embryonic stem cells and the apply RNA-seq technology to dig the profile of transcriptome in each part. Combined with public epigenome data and my previous whole cell transcriptome data, we will focus on the transcripts distribution and significance located on the inside and outside of the nucleus, differences between different types of transcription of this genomic characteristics, and distinguish the real function transcripts from transcriptional noise under the complex background. Our research will provide a solid basic foundation for transcription regulation research and the simulation of cellular regulatory networks in the future.
