中文摘要: 微RNA在各种疾病的形成过程中都起到了至关重要的作用。我们在本项目中不仅将寻找疾病与其之间直接二元关联,而且计划从其上下游寻找疾病形成深层次原因。从上游来看,环境因子会导致微RNA出现问题,从下游看之,微RNA对靶基因的调控会出现问题,这两种方式都会导致疾病形成。本项目研究内容包括三部分:(1)疾病与微RNA关联预测,解决哪些微RNA导致疾病发生这一问题;(2)疾病相关环境因子和微RNA作用预测,解决哪些环境因子导致微RNA出现问题从而导致疾病这一问题;(3)疾病相关miRNA-靶基因作用预测,解决微RNA对哪些靶基因调控出现问题导致疾病这一问题。从高通量生物数据预测潜在疾病相关作用是生物医学和生物数学中的重要问题。本项目将克服现有研究中存在的缺陷,从半监督学习、网络算法、数据结合等出发多视角建立数学模型与方法。本项目将为生物相关领域提供新的研究方法,并将推进对人类复杂疾病的深入理解。

英文摘要: MicroRNAs (miRNAs) play critical roles in the development and progression of various diseases. This project will identify not only the binary relationship between disease and miRNA, but also the depper causes of disease formation from the upstream and downstream of miRNA. From the upstream of miRNA, environmental factor would functionally interact with miRNA and their interactions would contribute jointly to diseases; From the downstream of miRNA, the dysregulation of miRNA-target interaction would also contribute to disease. This project will study the following three key scientific questions: (1) predict disease-miRNA associations, i.e. determine which miRNAs will cause the disease; (2) predict disease related miRNA-environmental factor interactions, i.e. determine which environmental factors would interact with miRNA and the interaction further contribute to disease; (3) predict disease-related miRNA-target interactions, i.e. determine which dysregulations of miRNA-target interaction would cause disease. Predicting potential disease-related interactions from vast amount of biological data and multiple angles is an important problem in the biomedical and biomathmatic research.
