中文摘要: 扩张型心肌病(扩心病)病因未明,越来越多的证据表明遗传因素在其发病中扮演了重要角色。我们前期基于新一代全外显子测序对一个扩心病家系进行分析,发现一个新的扩心病致病基因NOL8,表现为常染色体隐性遗传规律。目前国内外均未见该基因突变的报道。申请人前期研究工作表明:敲低斑马鱼NOL8的表达,导致斑马鱼胚胎心脏形态及功能异常,并出现与人类扩心病类似的特征。本项目拟利用敲除、敲低与过表达NOL8扩心病模型,研究NOL8与扩心病致病基因α-心肌肌动蛋白是否相互作用,以及其是否通过p38 MAPK信号通路调控扩心病的发生,从而阐明NOL8基因突变导致扩心病的病理生理机制。

英文摘要: Despite aggregating evidences demonstrate that genetic risk factors are involved in the pathogenesis of dilated cardiomyopathy, little is known about the underlying etiology in overwhelming majority of patients. A new NOL8 gene mutation was identified by analysis of next-generation exome sequencing data in a dialted cardiomyopathy family with an autosomal recessive inheritance pattern. Abnormal cardiac morphology and function, especially the sign similar to human dilated cardiomyopathy, were found in the zebrafish embryo after NOL8 was knocked down in our latest study. Therefore, p-p38/p38 signal pathway and the interaction between NOL8/NOP132 and alpha-cardiac actin, a pathogenic gene of dilated cardiomyopathy, will be investigated by the NOL8 knockout, knockdown and overexpression dilated cardiomyopathy zebrafish model. Accordingly, the involved pathophysiological mechanism of dilated cardiomyopathy will be expected to elucidation.
