中文摘要: RyhB是目前己知调控细菌靶mRNA数目最多的sRNA,项目组在前期成功构建禽致病性大肠杆菌(APEC)基因缺失株△RyhB及回复株△RyhB-comp并证明RyhB的基因缺失可引起APEC多个关键毒力因子差异表达的研究基础上,依据RyhB是APEC致病性调控网络关键点的新思路,以上述两株菌及亲本株为研究对象,应用二维电泳、高通量测序等技术,比较三株菌生物学特性的差异,筛选RyhB调控APEC的下游靶点(靶基因/蛋白)并预测调控通路;继而建立三株菌体、内外感染鸡气管上皮细胞的实验模型,比较细胞损伤差异,筛选差异表达的感染细胞应答基因并预测调控通路,该研究结果将为揭示RyhB调控APEC致病性的分子机理提供研究支撑。

英文摘要: RyhB is known currently not only as a kind of sRNA which may regulate the largest number of bacterial target mRNA but also as a key component of bacterial regulatory networks. Base On constructing the RyhB gene-deleted APEC strains(△RyhB) and gene-replied strains(△RyhB-comp) and according to the new idea what RyhB is a key point of APEC regulatory networks, we wound reseach the two strains and parent strains as follows using analyze technology such as 2D-electrophoresis, HTS and so on. Firstly we may compare of bionomics among three strains and then screen the gene/protein targets in APEC regulated by RyhB. Secondly we wound construct the experimental modle of chicken trachea epithelium cells (CTECs) infeced by the three strains in vivo or in vitro, respectively and compare of the damage differences among the cells. Firstly we wound screen the .response Gene of the infected-cells effected by RyhB.
