中文摘要: LRR-only蛋白是近年来备受关注的一类新型模式识别受体,已有研究进展提示该蛋白家族在固有免疫防御机制中发挥着重要作用。目前对海洋无脊椎动物LRR-only蛋白免疫功能的报道几近空白。本项目拟在前期获得3种栉孔扇贝LRR-only蛋白基因的基础上,综合运用各种现代生物学技术手段,从栉孔扇贝中进一步发掘LRR-only蛋白家族成员,分析其序列的保守性和多样性;在个体或细胞水平研究这些分子的表达调控规律;体外重组这些蛋白,筛查其识别和结合配体的种类及结合活性;向原代培养的扇贝细胞中添加这些基因的重组蛋白或通过RNA干扰沉默这些基因的表达,检测这两种处理对各类下游免疫指标的影响;综合以上研究结果阐释该蛋白家族对机体免疫应答的调控作用。本项目的研究成果将为深入了解软体动物LRR-only蛋白的免疫调控作用提供有力证据,丰富和扩展海洋无脊椎动物免疫学内容。

英文摘要: The LRR-only proteins have been confirmed to be involved in innate immune responses as a new kind of pattern recognition receptor (PRR) and might play a pivotal role in the innate immune system. However, the mechanism of immune regulation mediated by LRR-only proteins in invertebrate innate immune system is still unclear. In our previous research, three LRR-only protein genes have been identified in Zhikong scallop Chlamys farreri and one of them has been investigated preliminarily. Based on such achievements, the present research will be focused on the full-length cDNA cloning and sequence feature analyzing of more LRR-only proteins by molecular technique and bioinformatics software. The mRNA distribution and expression profile of these genes will be observed via real-time PCR and their binding activity to various ligands will be tested through ELISA based ligand-binding assay or ProteOn XPR36 protein interaction system. RNAi and more other methods will be employed to investigate the immunomodulation mechanism of LRR-only proteins. The results of our research will reveal the mechanism of immune regulation mediated by LRR-only proteins in mollusk and enhance the understanding of the immune defense system in invertebrates.
