中文摘要: Th17细胞在病原体诱导的慢性炎症过程中发挥重要作用,但在慢性乙型肝炎肝损伤中的作用及调控机制尚不明确。我们前期研究发现,Th17细胞参与慢性乙型肝炎肝脏的炎症损伤过程。同时,在慢性HBV感染,病毒抗原不仅诱导机体产生Th17细胞免疫,而且刺激免疫抑制因子IL-10的分泌增加,IL-10则可抑制HBV抗原特异性Th17细胞的分化。据此推测:慢性HBV感染宿主通过促进抑炎分子IL-10的分泌,减轻Th17细胞对肝脏强烈的免疫损伤作用。本课题拟采用流体动力学法构建慢性HBV感染小鼠模型,应用RNAi干扰技术、抗体特异性阻断技术等,从细胞培养和动物体内两个层面研究IL-10调控Th17细胞分化在慢性HBV感染肝损伤中的作用;并基于JAK-STAT信号通路,研究IL-10调控Th17细胞分化的确切分子机制,为进一步阐明慢性HBV感染肝损伤的免疫发病机制,寻求更有效的肝脏炎症损伤干预靶点提供依据。

英文摘要: Th17 cells are important effector cells in the microenvironment of chronic inflammation and play an important role in chronic inflammation induced by pathogens. Howeever, the role of Th17 cells in chronic hepatitis B liver injury and the modulate mechanism is unclear. Our preliminary study found that Th17 cells involved in the process of inflammatory liver injury induced by hepatitis B virus. Then further in vitro study confirmed that IL-10 inhibits antigen-specific Th17 cells differentiation in patients with chronic hepatitis B. Thus we put forward the hypothesis of this subject: HBV induces inflammatory Th17 cell immunity in the host of chronic HBV infection, at the same time promotes the anti-inflammatory molecule-IL-10 secretion to inhibit the strong liver immune injury through limiting Th17 cells differentiation. To this end, we use the fluid dynamics method to establish a mouse model of chronic HBV infection, then we use this model to explore the exact molecular mechanism of IL-10 regulate Th17 cells differentiation in chronic HBV infection through modulating JAK-STAT signaling pathway. Our findings may aid in the design of new immunotherapy for intervention of CHB in clinics.
