中文摘要: 针对马铃薯与青枯菌互作分子机制系统研究不足以及马铃薯抗青枯病育种缺少相应的分子病理模型的现状,本项目拟采用新的研究策略探讨马铃薯与青枯菌互作的分子机制。借鉴动植物病原生态位模型,应用CFU计数和环境扫描电镜检测技术鉴定根际生态位、根维管束生态位和茎部维管束生态位的建立情况,分别分离病原菌与寄主RNA,采用微阵列和SSH-EST技术分别检测青枯菌生态位特异的转录组和寄主相应的应答转录组(尤其是防卫相关基因表达谱),结合生物信息学等技术进行基因功能注释和分类,对产生的不同生态位特异的转录组进行比较分析。探索相关基因在二者互作中的相互影响,以期系统地了解生态位特异的转录组中基因的种类、功能和表达模式,推断生态位特异表达基因在马铃薯与青枯菌互作中的作用。对于阐明马铃薯与青枯菌互作的分子机制有重要意义,为进一步针对马铃薯抗病分子育种提供选择相应的育种策略奠定一定的理论基础和提供新的思路。

英文摘要: Very little is known about the molecular mechanism of potato-Ralstonia solanacearum Interactions. It's insufficient to have some corresponding molecular pathology models to guid the resistance breeding aganist bacterial wilt in potato. Our project provides the new research strategies for researching the molecular mechanism of interaction. By referencing the current situation of animal and plant pathogen niche model, applying of the CFU count and environment scanning electron microscopy (ESEM) testing technology to identify the establishment of the ecological niche for the baterial colonization,the RNA will be extracted for constacting the transcriptome pfrofiles using the microarray and SSH-EST technology in both the pathogen and the plant host. And the comparative transcriptmics analysis will performed with the methods of bioinformatical technology and function classification to explore the related genes for their types, functions and expression partterns that would be hlepful to analyze the possible mechanism between poato and pathogen based on the different pathogenic niches in which there must be niche-specific gene expression in the pathogen and corresponding gene expression in the potato host.
