中文摘要: 病害问题一直是影响对虾养殖业可持续发展的最大障碍,且对虾白斑杆状病毒(WSSV)是被认为对虾养殖业危害最为严重的病毒。前期的研究结果证实,NF-κB通路、JAK/STAT 通路以及抗菌肽等免疫效应因子在对虾免疫防御WSSV过程中具有重要的作用,因此系统地研究这些免疫相关基因的SNP位点与对虾抗WSSV性状的关系,鉴定抗病相关SNP位点并解析抗性机理,对于辅助抗病品种的培育具有重要的意义。本研究拟在课题组已获得的凡纳滨对虾大量转录组数据和基因组数据的基础上,通过生物信息学方法全面分析参与WSSV免疫防御过程中的信号通路和效应因子基因中SNP位点,利用关联分析方法确定与对虾抗WSSV性状关联的SNP位点,进一步分析抗性相关SNP导致的蛋白变化以及调控基因的表达变化等,对其抗性差异的机制进行初步分析。研究结果可为对虾抗性性状的解析及其辅助抗病品种的培育奠定重要基础。

英文摘要: Diseases have been the biggest obstacle to the sustainable development of shrimp farming and White Spot Syndrome Virus (WSSV) is considered the most serious virus affecting the shrimp farming industry. Previous study have proved that NF-κB pathway, JAK/STAT and immune factor like ALF play important roles in response to WSSV infection. Therefore, It is of great significance to investigate the SNPs in these immune gene and investigate the mechanism of WSSV resistance caused by SNPs. Based on the transcriptome sequencing and genome sequencing data obtained previously, this study will focus on the development of large amounts of SNP markers in immune pathway and immune factor genes by bioinformatics method. Then, the association analysis will be conducted to screen WSSV resistance SNPs. The mechanism of resistance caused by the SNPs will also be conducted by analyzing protein and gene expression change of immune genes. From this study, it will provide important basic for elucidating disease resistance trait and assisting disease-resistance breeding.
