中文摘要: 本研究利用DNA"分子机器"的信号放大机制,以可卡因为研究对象,设计高灵敏生物检测方法。在前期工作中,我们利用核酸适配体识别可卡因,设计了一种仿PCR型的DNA分子机器,利用其对核酸信号的放大功能,对可卡因的检测灵敏度可以达到微摩级别。本项目拟采用两种DNA分子机器,分别为多重循环仿PCR型分子机器和滚环放大型DNA分子机器,深入研究理解DNA分子机器在传感器应用中的基本物理、化学问题,在此基础上进一步提高对可卡因的检测灵敏度,同时尝试对实际样品中的可卡因检测。本项目拟采用凝胶电泳、原子力显微镜、荧光光谱等表征手段,在对这一体系得到规律性认识的基础上,将DNA分子机器推广为适用于其他对象的传感方法。

英文摘要: In this proposal, We propose to design novel DNA machine based cocaine detection approach. In our previous work, we have developed a PCR mimicking DNA machine using anti-cocaine aptamer as a capture probe, and a detection limit as low as uM could be easily achieved. In the present project, we will design two novel DNA machine based cocaine detection approaches, which are cascaded PCR-mimicking machine and rolling cycle amplification machine, respectively.These will help us to deepen our knowledge of the detailed physical and chemical mechanism of the DNA based machine, which would be applied for further cocaine sensors with improved sensitivity. In addition, we will also challenge the present sensors with real samples. Gel-electrophoresis, AFM and fluorescence measurements will be used in this study. Once we obtain a general understanding of this system, this DNA machine based detection strategy will be further applied for other sensing system.
