中文摘要: 随着我国新鲜水果消费的种类和数量大幅增加,水果已经成为我国临床上发现的主要致敏食物,桃是我国最常见的大宗过敏水果之一,影响部分人群的安全和生活质量。根据前期临床免疫学研究表明,我国95%桃过敏患者是对一种脂质转移蛋白(过敏原标准命名为Pru p 3)过敏,引发较为严重的系统性过敏反应。解决桃过敏问题的一个有效方法是鉴定选择低过敏的品种。本项目将分离纯化天然的桃果实脂质转移蛋白,进行酵母表达系统重组生产,制备单克隆抗体,应用ELISA方法对90多份桃核心种质材料进行过敏蛋白的定量分析,比较不同品种类型和起源等与过敏蛋白含量的关系,并进行全基因组SNP关联分析其遗传控制机理,开发相应的分子标记应用于选择低过敏桃品种。

英文摘要: With dramatic increase of diverse fruits consumption in China, fruits have been recognized as the number one food allergy in clinics that affect patients' safety and quality of life. Peach is frequently reported as allergenic fruit and recently we identified the lipid transfer protein (Pru p 3) is the dominant major allergen causing severe systematic symptoms. Also significant differences in the gene expression were demonstrated between various peach cultivars, so selection of low Pru p 3 content is a positive preventive strategy to peach allergy problem. This project will isolate natural Pru p 3 protein and produce recombinant allergen by yeast expression system. Monoclone antibody will be prepared and used in measuring Pru p 3 content of 90 diverse peach culitvars. Moreover, genome wide association analysis using high denisty SNP will be performed to identify specific loci that allow breeders to select hypoallergenic cultivars.
