中文摘要: 镉污染已成为世界范围内影响较广泛、危害较大的环境问题之一。狗牙根具有发达的根状茎, 能够形成致密的地表覆盖,可有效减少镉污染土壤因雨水冲刷所造成的镉流失和扩散。不同生态环境野生狗牙根种群存在广泛的遗传变异,但对不同材料间耐镉性以及分子遗传基础缺乏深入的研究。本项目应用不同生态环境野生狗牙根种群为材料,以栽培品种作为对照,在温室可控条件下进行不同镉胁迫处理,研究:(1)不同生态环境野生狗牙根种群与耐镉性状相关的生长和生理指标差异,构建狗牙根耐镉评价体系;(2)通过关联分析筛选与狗牙根耐镉基因位点连锁的分子标记位点,并明确不同耐镉基因位点效应的大小;(3)采用极端耐镉和对镉敏感狗牙根杂交后代材料,验证耐镉基因的检测位点。通过本项目的实施,以期阐明狗牙根耐镉的分子遗传基础,为狗牙根耐镉品种的育种提供理论和实践上的依据。

英文摘要: Cadmimum is one of the most pollutants we are facing. Bermudagrass (Cynodon spp.) is an important warm season grasses widely used in China as turfgrass, forage grass, and phytoremediation. Bermudagrass covered the soil surface with dense rhizome root and shoot and prevent Cd polluted soil from erosion. However, the present bermudagrass cultivars are usually not good at Cd tolerance. Wild bermudagrass is rich in stress resistance gene and can be used for the genetic improvement of bermudagrass cultivars as precious germplasm resources. However, the divesity in Cd toelrance and molecular mechanism among wild bermudagrass resouces have not been docummented well. We intend to screen the wild bermudagrass with great Cd resistance from 404 accessions of germplasm and to explore new Cd resistance gene loci. The objectives of this study is to i) evaluate the diference in Cd tolerance of bermudagrass accessions and establish an evaluation method of Cd tolerance; ii) examine the gene alleles related to Cd tolerance of bermudagrass via association analysis and assess the effects of them; iii) verify the candidate Cd alleles using F1 population of Cd toelrance accession Cd sensitive accesion. This study will improve the understanding of the Cd-resistant molecular mechanism in wild bermudagrass and promote the use of genetic resources of wild bermudagrass in Cd resistant breeding
