中文摘要: 芝麻枯萎病是一种分布广、防治困难的世界性真菌病害,严重威胁和阻碍着芝麻的生产与发展。由于抗病过程较为复杂且往往涉及复杂的网络系统,这给抗性基因的挖掘带来困难。以连锁不平衡为基础的关联分析方法的完善以及大量芝麻分子标记的成功开发,为芝麻枯萎病抗性与基因的关联分析研究提供了前提条件。本项目拟首次用全基因和候选基因关联分析法研究芝麻枯萎病抗性:拟构建250份用于关联分析的自然群体,并对其进行枯萎病抗性鉴定。利用基于高密度芝麻遗传图谱上的大量genic-SSR标记以及转录组分析获得的18个抗性候选基因SNP标记,对该群体进行全基因组扫描和群体结构分析;随后利用关联分析方法对表型性状和分子数据进行关联,获得与抗病相关的优异等位基因,并在表达水平方面对其进行进一步的功能验证。该究将为抢占芝麻抗性基因资源、提升芝麻基础研究水平和利用分子手段培育高抗芝麻新品种奠定基础。

英文摘要: Sesame Fusarium wilt was a common and hardly controlled fungus disease all over the world, which could cause economic losses seriously on sesame. Due to the complex network system between the resistant traits and genes involved in disease resistance, it is difficult to discovery the resistance genes. Fortunately, the development of association analysis method based on linkage disequilibrium (LD) and application of vast molecular markers in sesame genetic analysis, offer a new tool for the association analysis between specific loci and this trait. In this study, it is the first time to study the resistance of sesame Fusarium Wilt by genome-wide and candidate gene association analysis. A systematic resistant identification would be performed in 250 sesame germplasm resources. Then the genome-wide scan, genetic diversity, population structure and the LD analysis of 250 sesame germplasm resources would be assessed by a mass of genic-SSR markers from a fine sesame genetic mapping and 18 candidate resistant gene SNP markers from transcriptome analysis. Finally, the Fusarium wilt resistant association analysis and qRT-PCR methods would be chosen to verify the candidate resistant gene.
