中文摘要: 黄瓜再生体系作为离体快繁和遗传转化的基础对黄瓜生物技术研究和种质资源创新都具有重要意义。特别是随着黄瓜基因组测序的完成及功能基因组学的开展,更需要稳定、高效的再生体系提供技术支撑,但目前制约其发展的最严重问题依然是再生频率低、再生体系适应性差。可见黄瓜不同品系之间在再生能力上存在着明显差异,但遗传机理又尚不明确,因此对黄瓜再生能力差异进行研究显得尤为重要。本项目拟在利用两份已经测序完成的黄瓜材料构建的RIL群体为基础,研究子叶再生能力差异的遗传规律,并进行基因定位;利用109份黄瓜重测序材料,通过全基因组范围内的关联分析发掘与子叶再生控制基因紧密关联的SNP,寻找相关的基因位点。本项目将从分子遗传学角度解读基因型间再生差异的成因,为建立快速、高效、稳定的黄瓜子叶再生体系提供理论依据和实用信息。

英文摘要: As the basis of in vitro rapid propagation and genetic transformation of cucumber regeneration system the cucumber biotechnology research and germplasm resources of innovation are of great significance, especially with the completion of the cucumber genome sequencing and the development of functional genomics, also needs a stable, efficient regeneration system to provide technical support, but the most serious problems currently restricting its development is still low regeneration frequency regeneration system adaptability. There are significant differences in regenerative capacity between different strains of cucumber, but the genetic mechanism is not clear. Its launches research to the cucumber reproductive property difference appears especially important. In this project, the inheritance and gene mapping of cotyledon regeneration differences will be studied using a RIL population derived from the cross of two sequenced cucumbers. 109 cucumbers are referencing materials, through genome-wide association study to mine gene closely related to the cotyledon regeneration, look for associated loci.
