中文摘要: 大豆异黄酮在植物防御系统中发挥重要作用,并且与人类健康密切相关,提高大豆异黄酮含量是重要的育种目标之一。大豆异黄酮通过植物共有的苯丙氨酸代谢途径和豆科特异的异黄酮分支途径合成,受多基因和复杂网络调控,过表达途径上的个别基因通常对提高大豆异黄酮含量效果不显著。只有对途径上的多基因进行整体分析,才能有效揭示大豆异黄酮含量的遗传控制。本项目以大豆异黄酮代谢途径基因为研究对象,通过大豆物种内和近缘物种间遗传差异分析,探索异黄酮代谢途径基因的进化模式。然后分别对单个候选基因进行关联分析,发掘与大豆异黄酮含量显著相关的SNP标记和优异单倍型。进而通过多基因关联分析,了解基因间的相互作用方式,推测优异等位基因组合。本研究将从新的视角阐述大豆异黄酮含量的多基因遗传控制,并为提高大豆异黄酮含量、培育高异黄酮含量品种提供基础。

英文摘要: Given the positive effects on plants and human health, increasing isoflavone contents is one of the targets of soybean breeding programs. Isoflavones which are synthesized through the central phenylpropanoid pathway and the specific isoflavonoid branch pathway in legumes are controlled by a complex network of multiple interactive genes. Overexpression of certain genes on the pathway could not increase the isoflavone contents significantly. Overall consideration of multiple genes on the pathway will not only deepen the understanding of the genetic control of isoflavone content, but also provide a reference for other complex traits and pathways. This study will focus on genes of the isoflavone biosynthetic pathway. The evolutionary patterns of isoflavone pathway genes will be surveyed by intra- and inter-species genetic variation analysis. Significantly associated SNP markers and favorable haplotypes will be identified using candidate genes association analysis. Through multigene association analysis, the interactions among genes and elite allele combinations will be presumed. In conclusion, we will discuss the multi-genetic control of soybean isflavone content, and the potential for the improvement of soybean isoflavone content.
