中文摘要: 神经干细胞(Neural stem cells,NSCs)的分化调控机制是神经干细胞基础与临床应用研究中的关键问题。视黄酸(Retinoic acid, RA)是参与神经分化和神经形成的重要神经发育信号分子,可诱导NSCs向神经元分化,但其调控NSCs 分化过程中的蛋白磷酸化动态研究尚未见报道。本研究以RA诱导NSCs分化为模型,主要利用细胞培养稳定同位素标记SILAC技术为主的定量蛋白质组学技术进行蛋白磷酸化修饰的动态分析,结合生化/分子/细胞生物学及生物信息学等生物学手段,对RA诱导NSCs分化过程中的关键磷酸化蛋白进行深入功能分析,研究筛选的关键磷酸化蛋白在NSCs分化中调控RA信号中的RAR-RXR受体与其相关转录共激活与共抑制复合物,进而调节下游SOX1和MAP2等神经干细胞标志基因和神经元标志基因的途径。本研究对于探索NSCs分化机制和神经退行性疾病的治疗具有重要意义。

英文摘要: The regulation of neural stem cells (NSCs) differentiation is an important question in the basic research and clinical application of NSCs.Retinoic acid (RA) is a critical signaling molecule, which is involved in neural differentiation and neurogenesis. However, the molecular mechanism underlying RA-induced neural stem cell diferentiation remains unclear. In this study, we utilized the SILAC (Stable isotole labelling by amino acids in cell culture) quantitative phosphoproteomics technique to analyze the protein phosphorylation dynamics of RA-induced rat NSCs differentiation. In combination of various biogical techniques including biochmistry, molecular biology, cell biology and bioinformatics, we will examine the function and regulary mechanism of critical phosphorylated proteins in RA-induced NSCs differentiation.Furthermore, we will investigate the impacts and specific pathways of critical phosphoproteins on the regulation of RAR-RXR receptors, their related transcriptional coactivators and corepressors, and the downstream neural stem cell markers and neuron markers,such as SOX1 and MAP2.Our studies would provide many important clues in the investigation of the molecular mechanism of NSCs differentiation and the neurodegenerative therapy.
