中文摘要: 视网膜缺血-再灌注(RIR)损伤可以导致视神经节细胞(RGCs)死亡、视网膜形态退化,最终导致视力丧失,目前尚无有效的治疗方法。骨髓来源的神经干细胞(BM-NSCs)可以分化为神经节并且具有免疫调节的作用,IL-10可以抑制炎症反应,并且对神经干细胞(NSCs)生物学特性有一定的调节作用。目前BM-NSCs和IL-10对RIR损伤作用的研究严重缺乏。我们的前期研究已建立大鼠RIR损伤模型,并且研究了RGCs的凋亡情况和凋亡相关基因的表达变化。本项目中我们将进一步探讨以下问题:(1)IL-10对BM-NSCs生物学特征的影响;(2)IL-10修饰的BM-NSCs对RIR损伤的作用;(3)IL-10修饰的BM-NSCs对RIR损伤作用的下游分子机制的研究。通过本项研究,我们将明确IL-10修饰的BM-NSCs在RIR损伤中的作用及其分子机制,将为RIR损伤的临床治疗提供有力的理论基础。

英文摘要: Retinal ischemia/reperfusion (RIR) injury can lead to the death of retinal ganglion cells (RGCs), morphological degeneration of retina, the loss of retinal function and ultimately, the loss of vision. Bone marrow-derived neural stem cells (BM-NSCs) can differentiate into neuron and regulate the immunoreaction. IL-10 can inhibit the inflammation and regulate the phenotypes of NSCs. Currently, the ability to harness BM-NSCs as potential therapies in RIR injury is compromised due to the significant knowledge gap. Our group has established the RIR injury rat model and studied the apoptosis of RGCs cells. Our objectives are (1) the effect of IL-10 over expression on phenotypes and functions of BM-NSCs; (2) the role of IL-10-modified BM-NSCs in progression of RIR injury; (3) the molecular mechanism of IL-10-modified BM-NSCs in progression of RIR injury. We will conclude the function and molecular mechanism of IL-10-modified BM-NSCs in progression of RIR injury, and provide a rationale for developing therapeutic methods to treat clinical RIR injury.
