中文摘要: 蛋白质相互作用网络是系统生物学的核心研究内容,挑战是如何对大规模的网络数据进行分析和挖掘,以获取生物体的结构和功能信息,预测潜在的疾病模块。研究发现同一功能或者疾病模块内的所有细胞元件,极有可能参与相同的疾病,而且疾病模块具有保守性。因此,通过研究在蛋白质网络中挖掘功能模块,以及通过模块比对发现保守功能模块,能够发现潜在的疾病模块。本项目主要研究:(1)从复杂系统的角度,研究蛋白质网络的拓扑结构特征。(2)基于系统的核与核度理论、图聚类分析和关联规则技术,通过识别蛋白质网络中的功能模块来发现潜在的疾病模块。(3)通过不同物种功能模块间的比对分析,发现保守功能模块来预测疾病模块。(4)对疾病模块进行深入分析,试图理解疾病的分子机制, 发现可能导致疾病的原因,为疾病的医治提供新的治疗方向。

英文摘要: Protein-protein interaction networks are the core research contents of system biology.The challenges are how to analyze and mine the large-scale network data to obtain the structural and functional information of the organism,and predict the potential disease modules. Evidence from many sources suggests that all cellular components that belong to the same functional or disease module have a high likelihood of being involved in the same disease.And a conserved module is likely to be a disease module. Therefore, by identifying functional and conserved functional modules from protein interaction networks, we study algorithms for mining disease modules.The main contents of this research project include:(1)From the perspective of complex systems, we study the topological attributes of protein networks.(2)Based on the theory of system core and coritivity,cluster analysis and association rule technology,we identify functional modules to find candidate disease ones from protein networks.(3)By the comparative analysis between functional modules of different species,conserved functional modules are detected to predict disease ones.(4)We also make a detailed analysis of disease modules to discover the potential molecular mechanisms of disease, and provide new directions for disease therapy.
