中文摘要: 自相互作用蛋白质指的是某种蛋白质的两个或者两个以上拷贝能够发生相互作用的蛋白质。自相互作用蛋白质对细胞发挥功能以及生物网络进化都起到重要作用。然而目前最常用的两种检测蛋白质相互作用的高通量实验技术- - 酵母双杂交和亲和纯化质谱在检测蛋白自相互作用方面的局限性可能导致人们大大低估了自相互作用蛋白的数量。本课题首先从序列、功能、进化、网络拓扑和基因表达等多方面对自相互作用蛋白展开系统研究,以系统揭示自相互作用蛋白的特点,而后基于其在多方面的特点,发展一个自相互作用蛋白质预测体系,最后给出一个注释详细、数据全面的自相互作用蛋白集合(包括有实验证据支持的和预测的自相互作用蛋白数据)。该工作不但有助于系统理解自相互作用蛋白在细胞功能中的作用,同时所发展的首个自相互作用蛋白预测体系也将有助于自相互作用蛋白的高通量发现,为解读蛋白质功能提供线索。

英文摘要: A self-interacting protein is referred to as the protein whose two or more copies can interact with each other. Self-interacting proteins play an important role in cellular function and in the evolution of biological networks. The current two most common high-throughput assays used to detect protein interactions, yeast two-hybird (Y2H) and affinity purification with mass spectrometry (AP-MS), have limited ability to discern protein self-interactions, which may lead to the underestimation of the number of self-interactions. This project systematically analyzes self-interacting proteins from multiple aspects including sequence, function, evolution, network topology and gene expression, and then based on these properties, develops a self-interacting protein prediction system. Finally, this project will give a self-interacting protein dataset together with abundant annotation information, including known and predicted self-interactions. This project may help us systematically understand the cellular function of self-interacting proteins, and the first self-interacting protein prediction system can contribute to the high-throughput finding of self-interacting proteins and provide clues for elucidating protein function.
