中文摘要: 本课题的目的在于开发基于微流控平台的单细胞电化学分析新方法,实现单细胞水平、高通量、多参数、快速可靠的芯片上电化学分析。将其与芯片上细胞连续培养、环境控制相结合,实现对群体细胞中的细胞亚群的高精度的识别和分选。通过本课题的研究,我们旨在(1)利用微系统局域特点进行电化学单细胞分析,降低分析的检测限,利用微流体的精确操控能力实现同一电极上多种物质的检测,进而获得单细胞水平各种表达物的关联信息;(2)构建具有上述功能的高通量电极阵列,获得单细胞水平群体细胞的统计信息,为单细胞水平癌症细胞的分选和癌症干细胞的识别提供依据。

英文摘要: In this project, we aims to develop powerful single cell electrochemical analysis methods based on microfluidic platforms, by which the analysis can be high throughput, multiplex, fast and reliable. Combing these characters with ability of on chip continuous cell culture, controllable environment stimulation, the platform would enable concise sorting and recognition for sub-populations from whole population of cells. By carrying out this project, we aims to (1) manipulate the advantage of microscale confinements for electrochemical single cell/clone analysis, reducing the detection limit; take advantage of the accurate controlling ability of microfluidics for multiplex electrochemical analysis on a same electrode, and thus get the multiplex information from a same cell; (2) buildup highly assembled electrode array with the above mentioned functions for statistical information of single cells in a cell population, and thus facilitate cancer cells sorting and CSCs recognition.
