中文摘要: 大肠癌严重危害我国人民身体健康。申请者前期研究发现,临床应用复方肠泰能够改善大肠癌患者临床症状,降低肿瘤标记物的水平,并且提高T淋巴细胞分型为代表的多项免疫指标。同时,亦发现复方肠泰能够诱导大肠癌细胞自噬水平提高。但复方肠泰是通过何种机制提高免疫功能并发挥其抗肿瘤效应,尚不清楚。. 抗肿瘤免疫应答的核心是肿瘤细胞免疫原性,而肿瘤细胞释放的自噬小体与肿瘤细胞免疫原性密切相关,肿瘤细胞来源的自噬小体可能是肿瘤抗原的有效载体。为此,本项目将在前期研究基础上,以肿瘤细胞自噬小体的抗原提呈功能为切入点,将复方肠泰诱导大肠癌细胞自噬与肿瘤细胞免疫原性及特异性抗肿瘤免疫应答相联系,从肿瘤免疫学原理来阐明复方肠泰的抗肿瘤作用机制。为中药在抗肿瘤免疫应答这一肿瘤研究热点领域的作用提供实验结果和理论依据。

英文摘要: Colorectal cancer seriously harm people's health in China. Our previous studies found that clinical application of Fufangchangtai decoction can improve clinical symptoms in patients with colorectal cancer, reduce the levels of tumor markers, and increase the T lymphocyte numbers. Meanwhile, we also found that Fufangchangtai can induce autophagy of colorectal tumor cells. However, the mechanism of Fufangchangtai improve the immune function is not clear.. The core of anti-tumor immune response is the immunogenicity of the tumor cells. Autophagosomes released by tumor cells is closely related to the tumor immunogenicity. Tumor-derived autophagosomes may be an effective carrier of tumor antigen. Therefore, based on the previous research, this study will focus on whether autophagy of colorectal tumor cells induced by Fufangchangtai can enhance tumor immunogenicity by cross-presentation and activate specific anti-tumor immune response. This study could provide experimental results and theoretical basis to research of traditional Chinese medicine affecting the antitumor immune response.
