中文摘要: 虹彩病毒是石斑鱼等主要海水养殖鱼类的重要传染性病原,危害极大,然而,目前对这类病毒在鱼类宿主细胞内的精细侵染机制仍不清楚。本项目拟在前期工作的基础上,通过交叉运用生化、分子生物学和生物物理学等技术,在单病毒粒子水平系统开展石斑鱼虹彩病毒SGIV侵染宿主细胞的精细机制研究。在活细胞内直观揭示SGIV病毒进入宿主细胞的方式、途径和精细动态过程,阐明其入膜和跨膜入侵机制;从时间、空间尺度实时动态监测SGIV病毒在复制和运输过程中与细胞骨架成分的相互作用,分析其运输途径、运输过程和动力学特征。本研究首次将单病毒粒子示踪技术应用于鱼类大分子DNA病毒研究,研究结果将从新的角度进一步阐明鱼类虹彩病毒感染致病的细胞基础和分子机制,同时为有针对性地发展鱼类病毒病防控技术奠定基础。

英文摘要: Iridoviruses are one of the most important viral pathogens in culture groupers and other marine fishes. However, little is currently known about the infectious entry process of iridovirus into host cells. Based on the previous studies, we will thoroughly investigate the detailed infection mechanism and intracellular motility of Singapore grouper iridovirus (SGIV) in host cells at single-virus particle resolution using biochemistry, molecular biology and biophysical technology. Firstly, we will disclose the pathway of virus entry and the detailed dynamic process. Then, we will investigate the interaction between the virus and the cytoskeleton components during SGIV replication, and analyze the dynamic process during virus movement. In this study, the single-virus tracking will be firstly applied to fish large DNA virus. Our results will not only provide new insights into elucidating the molecular mechanism of fish iridovirus disease, but also contribute greatly to developing novel and effective strategy to preventing fish viral disease.
