中文摘要: 鸭肠炎病毒(DEV)是危害养鸭业的重要病原之一,但对该病毒的研究多集中在病毒基因组和蛋白质组上,极少涉及到病毒蛋白受体。RNA干扰技术是近10年发展起来的一门技术,已广泛应用到基因功能研究、抗病毒感染和肿瘤疾病治疗等多个领域。项目申请人在前期工作中,通过酵母双杂交系统筛选得到了DEV核衣壳蛋白(NP28)的宿主细胞受体即蛋白激酶C抑制蛋白(PKCI)。本项目拟在此基础上,应用RNA干扰技术分别对宿主细胞PKCI及PKC基因和病毒NP28基因进行沉默后,采用实时荧光定量PCR、电子显微镜、Western blot和免疫染色法等手段,研究分析PKCI及PKC基因表达与DEV增殖的相关性,明确PKCI及PKC在DEV增殖中的作用,以期为DEV致病机理的阐明和抗病毒药物的开发奠定基础。

英文摘要: Duck enteritis virus (DEV) is one of the major pathogens which danger to duck industry. At present, the studies for DEV is more concentrated on its genome and proteome, while seldom related to the receptor of viral protein. RNA interference (RNAi) is a technology developing in nearly 10 years and applied widely on the study of gene function, treatment of neoplastic diseases, defense of virus infection and other fields. In our preliminary studies, Protein kinase C inhibitor (PKCI) is confirmed as the receptor of DEV nucleocapsid protein (NP28) in the host cell by the yeast two-hybrid system. The objective of this study, following our previous studies, is to confirm the role of PKCI and PKC on the proliferation of DEV by methods of real-time PCR, Western blot, electron microscopy and immunohistochemical staining, after the PKCI gene, PKC gene and NP gene was silenced respectively by RNA interference. These studies may lay the foundation for elucidation of pathogenic mechanisms and development of therapeutic drug.
