中文摘要: 解除禽白血病病毒(ALV)的免疫抑制作用是攻克禽白血病的关键所在。前期研究中,我们发现,ALV的免疫抑制和病毒跨膜蛋白(TM)内高度保守序列密切相关。但其表达的小肽在机体内是如何行使其功能的?这一ALV诱导机体免疫抑制的关键问题,目前还不得而知。而外源体在这一过程中可能起到了至关重要的作用。本课题以ALV肿瘤细胞、免疫细胞分泌的外源体(exosome)为研究对象,以病毒学、免疫学前沿技术为手段,检测不同细胞来源外源体的生物学功能,通过对正向调控外源体及负向调控外源体单抗逆转ALV造成细胞及机体免疫抑制的研究,建立外源体正向调控和负向调控理论;以免疫细胞外源体、TM及骨髓树突细胞为基质,探索ALV疫苗研制的主要手段、方法及策略。本项目的完成,可以阐明exosome在ALV诱导免疫抑制中的作用,并在此基础上研制抗ALV的新型疫苗,同时也为其它囊膜病毒的防治提供新的思路和方法。

英文摘要: The key to control avian leukosis(AL) is get rid of the immunological suppression caused by avian leukosis virus(ALV). In many studies,we found that the immunological suppression is closely related with the conserved sequence of tranmbrane protein(TM).Up to now,we don't know the theory of the peptide in vivo.However, exosomes plays a vital role in the theory.This research project aims at discovering the theory of positive regulation and negative regulation, according to the study of the exosomes from different cells.We could seeking for apporaches and strategies of ALV vaccine development, on the basis of the exosomes from immunological cells,TM and dendritic cell.Once this project was finished,we could discovering the role of exosomes in immunological suppression caused by ALV, at the same time,this study will provide a new type of vaccine for ALV, and it will be a new opinion and approach of proventing other vriuses.
