中文摘要: 牛乳β-乳球蛋白形成低聚体是鲜牛乳中的一种自发现象,它对牛乳过敏性的影响尚未引起足够的重视。因此,本项目拟以天然存在的牛乳β-乳球蛋白低聚体为研究对象,分离并纯化β-乳球蛋白低聚体,进而采用动态光散射、电镜及光谱学等方法对其微结构和空间结构的特性进行表征。然后,通过模拟人工胃肠消化液,对β-乳球蛋白低聚体的热、酸和酶解的稳定性进行测定分析。最后,通过体外IgE结合能力和组胺释放能力的免疫学检测并结合体内的动物学实验进行评价,科学评估β-乳球蛋白低聚体的过敏性。 通过本项目的实施,将阐明牛乳β-乳球蛋白的结构变化与低聚体过敏性的关系,为控制β-乳球蛋白的过敏性提供新的理论依据。

英文摘要: The oligomers of bovine beta-lactoglobulin are presented in fresh milk, which not yet attracted sufficient attention to the impact on the milk allergy. Bovine beta-lactoglobulin oligomer was prepared by purification, and its microstructure and conformational structure were identified by dynamic light scattering,electron microscopy and spectroscopic methods. Then, the stability of beta-lactoglobulin oligomer was explored by heating, acid and enzymatic treatment by simulating artificial gastrointestinal digestive juice. At last, the allergenicity of beta-lactoglobulin oligomer was evaluated by immnological experiment of histamine release capacity and IgE binding ability in vitro and animal experiment in vivo. The work could probe the relation of structure change and the allergenicity of bovine beta-lactoglobulin oligomers, which hope to provide the theoretical basis for controlling the allergy of beta-lactoglobulin.
