中文摘要: 以鳞翅目害虫斜纹夜蛾为研究对象,以中肠专一表达的固醇转运蛋白SlSCPx为靶标,针对其重要结合底物胆固醇合成细胞膜和蜕皮激素20E影响幼虫生长发育的过程,利用脂质体转染技术在重组酶作用下将SlSCPx插入细胞的基因组,建立高效表达的SlSCPx蛋白的稳定细胞株,基于建立昆虫生长抑制剂高通量细胞筛选系统,对近万种化学合成小分子进行筛选,获得抑制胆固醇吸收的化合物。再经离体前胸腺离体培养和体外喂食实验等生物学验证和鉴定,以期获得有效的昆虫生长抑制剂。本项目通过建立昆虫生长抑制剂的高通量细胞筛选系统,获得有效的昆虫生长抑制剂,为进一步利用细胞高通量筛选技术获得有效的昆虫生长抑制剂提供实验素材和理论参考,对SlSCPx在害虫防治方面具有理论和现实意义。

英文摘要: In view of the important process of sterol carrier protein SlSCPx participating in cholesterol synthesis of ecdysteroid 20E and cell membranes to influence the growth and development of larvae of Spodoptera litura. This project will insert SlSCPx into the cell genome by using approaches of liposome transfection technique and finally acquire highly expressed SlSCPx stable cell line. Based on high-throughput cell-based screening system, nearly 10,000 kinds of synthetic chemical compounds will be screened. It can be further verified and identified by larval prothoracic glands cultured and the fed assay in vitro. It is hoped to obtain the effective insect growth inhibitors on pest control and lay a good theoretical for the further use of high-throughput cell-based screening system for effective insect growth inhibitors. It has important theoretical and practical significance for SlSCPx used on pest control.
