中文摘要: 大肠杆菌K1(E.coli K1)是引起新生儿细菌性脑膜炎及相关感染性疾病的主要病原菌,外膜蛋白A(outer membrane protein A, OmpA)作为其关键致病因子,在介导细菌引发宿主菌血症和入侵血脑屏障并引发感染性疾病过程中发挥重要作用。阐明细菌致病因子结构与其致病效应的关系是揭示细菌致病机制和治疗感染疾病的基础。课题组前期解析了OmpA的晶体结构,发现了E.coli K1的OmpA形成特异的结构域交互二聚体(domain swappeddimer)结构。为进一步揭示OmpA该特异结构在其致病效应中的作用,本研究拟通过同源重组、体外分析、细胞试验和动物实验等技术和手段分析该结构的变化对其介导的宿主菌血症和入侵血脑屏障作用的影响,以期阐明OmpA介导E.coli K1引发新生儿细菌性脑膜炎致病效应的机制,为研究E.coli K1新的治疗措施和药物靶点提供依据。

英文摘要: E. coli K1 is one of the major causitive pathogens of neonatal bacterial meningitis. As a key pathogenic factors of E. coli K1, Outer membrane protein A (OmpA) helps E.coli K1 induce host bacterimia and puncture the blood-brain barrier, which plays an important role in the infectious disease process. To clarify the relationship betweent the structure of bacterial pathogenic factor and the pathogenic effects is the basis of revealing the bacterial pathogenic mechanisms and treating infectious diseases. In preliminary study we determined the crystal strucuture of OmpA and found that OmpA forms a specific domain swapped dimer structure. In order to further clarify the relationship between the specific structure and the pathogenic effects of OmpA, we plan to answer the question by homologous recombination, cell and animal experiments, which help understand the pathophysiological pathogenic mechanisms of the bacterial neonatal meningitis caused by E. coli K1 and provide a theoretical basis for the study of new method and drug target for treatment of E.coli K1 infection.
