中文摘要: 大量的流行病学资料显示男性原发性肝细胞癌(肝癌)的发病率和死亡率均远远地高于女性,以往的研究认为其机制与性激素及其调控的信号通路有关,但以此为靶点的药物治疗收效甚微。前期的工作中我们发现定位于Y染色体上男性特异性区域的一个长链非编码RNA(命名为lncRNA-MSRY)特异性地在男性肝癌病人的癌组织中激活表达,而女性体内没有与之对应的基因组序列。预实验结果显示lncRNA-MSRY可促进肝癌的转移,其激活表达预示着肝癌病人的预后较差。本课题拟采用探针捕获深度测序、临床样品验证等手段对lncRNA-MSRY结合的蛋白、及其募集蛋白复合物至基因组特定位置的定位进行重点探究,揭示lncRNA-MSRY调控的下游效应分子和通路,并阐明控制lncRNA-MSRY表达的上游分子机制。为肝癌性别差异之谜的机制阐述提供新的角度,并为其治疗提供新的靶点。

英文摘要: Extensive epidemiological data show that male primary hepatocellular carcinoma (HCC) morbidity and mortality is much higher than female. Previous studies have mainly focused on the regulation of sex hormones and their pathways. Basing on these findings, people designed a number of drugs. But the effect is not ideal. This indicates that the male and female hormones difference is not the all the reasons for the gender differences of HCC prognosis. In previous work, we found that a long chain of non- coding RNA (named lncRNA-MSRY), located in a specific region of the Y chromosome (MSY) , was specifically expressed in male HCC tissue. Preliminary results showed lncRNA-MSRY may have an important role in the progress of HCC metastasis. This study focuses on the downstream effector molecules and pathways of lncRNA-MSRY and the molecular mechanisms lncRNA-MSRY upstream activation of expression. The purpose of the study is to find potential new drug targets for HCC therapy.
