中文摘要: 雌激素是动物体内产生的一类具有广泛生物活性的类固醇化合物,在雌性和雄性动物的生殖系统发育及生殖活动中都具有重要作用;但在雄性动物生殖作用研究中大多是从组织形态和机能变化方面开展,其作用机理尚不清楚。为此,本项目拟以贵州香猪为研究对象,二乙基己烯雌酚(DES)为研究材料,采用细胞培养、免疫组化、光学显微镜、 western blot,ELISA和实时定量PCR等技术,检测分析经DES处理后贵州香猪体内和体外培养的睾丸组织细胞(支持细胞、间质细胞和生殖细胞等)雌激素受体表达、一氧化氮合成酶活性及一氧化氮含量、Hsp90及SF-1表达和睾丸组织肥大细胞活性等的变化,以探讨雌激素在雄性动物生殖活动中的作用机理,这将有助于全面认识雌激素在雄性动物生殖活动中的作用,从而为改善动物繁殖性能和人类生殖健康提供理论依据和实践指导。

英文摘要: Estrogen, with a broad spectrum of biological activities, is a steroid compound hormone produced in animal's body; it has an important role in female and male animal reproductive system development and reproductive processes. The current data about the action of estrogen on male animal reproduction mostly focus on the tissue morphology and function change, but the mechanism of estrogen action is unclear. Therefore, in order to tackle the unclear question, Guizhou xiang male pig and diethylstilbestrol(DES) were used in the project, then cell culture, immunohistochemistry, optical microscopy, western blot,ELISA and real-time quantitative PCR technique were used for detection and analysis of estrogen receptor expression, nitric oxide synthase activity and nitric oxide content, Hsp90 and SF-1 expression and testicular tissue mast cell activity in the testicular tissue and/or testicular cells of guizhou xiang pig. The research will contribute to a comprehensive understanding the role and mechanism of estrogen on male animal reproductive processes, so as to provide the theory basis and the practice instruction for improving animal reproductive performance and human reproductive health.
