中文摘要: pVHL是一个经典的肿瘤抑制因子,它的突变和功能异常可导致肾脏透明细胞癌、嗜铬细胞瘤以及其它器官肿瘤的发生,但是对于其中的分子机制仍很不清楚。E2F1是一个重要的细胞周期调控基因,近年来也发现它在DNA损伤途径中发挥转录调控作用。我们在前期研究中,意外发现:pVHL不仅是一个受E2F1严格调控的基因,而且受到DNA损伤的诱导,此外,pVHL还与E2F1相互结合,并抑制E2F1的转录活性。这些初始结果不仅提示pVHL可能在DNA损伤途径中具有重要的新功能,而且还表明E2F1对VHL的调控可能是E2F1在DNA损伤途径中发挥作用的重要机制。本项目拟在前期工作的基础上,深入阐明pVHL在E2F1介导的DNA损伤途径和细胞周期进程中的作用及其分子机制,可望揭示pVHL在肿瘤发生和发展过程中的作用机制,为相关肿瘤的早期诊断、预防和药物筛选提供有益的理论指导。

英文摘要: VHL(von Hippel-Landau) is a classic tumor supressor, whose mutaions and dysfunction cause initiation and progression of clear cell renal cell carcinomas, pheochromocytomas and other tumours.However, the molecular mechanisms of pVHL in tumor suppression is still largely unknown. E2F1 is an important transcription factor in cell cycle control, which also have been implicated for acting its roles in DNA damage pathway. We accidently found that pVHL is a downstream gene of E2F1 and induced by DNA damage. In addition, pVHL interacts with E2F1 to inhibit its transactivity. These primiry results not only suggest that pVHL has a novel function in DNA damage pathway, but also indicate that E2F1 might act its role in DNA damage pathway by regulating VHL. Here, we propose to further investigate the role and molecular mechanisms of pVHL in DNA damage pathway and cell cycle progression which will reveal the cause of pVHL in tumore initiation and progression mechanistically and provide valuable guidance for diagnosis, prevention and drug screening of tumors.
