中文摘要: Small RNA是植物生长发育的一类重要调控因子。先期研究表明:small RNA可以在异源细胞之间移动与交流,并对受体的相关基因进行调控。因此我们推测,异源small RNA(不同种、属间)的移动与交流及其对受体基因的调控作用是远缘杂交中发生遗传变异的重要原因之一。本项目以芸薹属蔬菜(主要以榨菜与紫甘蓝为材料)的远缘无性与有性杂交为实验体系,利用qRT-PCR、MSAP、转基因等技术手段,进行异源small RNA的特异或差异表达、以及与性状变异相关基因的差异表达,甲基化模式分析及变异性状相关基因功能验证研究,阐明异源small RNA-基因-遗传变异之间的关系,明确异源small RNA诱导遗传变异的作用机制与模式,为农作物遗传改良和新种质的创制提供崭新的理论依据与途径。

英文摘要: Small RNA as a kind of key regulators play an important role in diverse aspects of plant development. In previous studies, the resules showed that small RNAs can exchange between heterologous cells, and regulate the expression level of genes in receptor. So we conjectured that the exchanges of heterologous small RNAs(different species)and its effection for the receptor is one of the important reasons for inheritable variation occurred in distant hybridization. Here we use the asexual and sexual progenies as experimental materials obtained from the distant hybridization in Brassica vegetables(Brassica juncea and Brassica oleracea ) and through the qRT-PCR、MSAP and transgenic technology to testify our hypothesis, and clarify the relationship of heterologous small RNAs-gene-morphological variation and define the molecular mechanism of inheritable variation regulated by heterologous small RNA through test the differential expression of small RNA and genes related characters variation, as well as analysis of methylation patterns between variant plants and control. The research will provides new methods for studying variation mechanisms induced by distant hybridization.
