中文摘要: 杂食性昆虫以其复杂的解毒酶系统,对不同毒性的植物次生物质进行解毒,而使昆虫得以进食多种植物;同时也会造成进食不同植物的昆虫对杀虫剂敏感性的不同。因此,这给以解毒酶为靶标开展害虫防治带来困难。目前昆虫解毒酶应答植物的表达调控途径研究极少,特别是在转录和转录后水平调控的研究未见报道。本项目针对从重要农业害虫斜纹夜蛾中发现的谷胱甘肽转移酶SlGSTE1,从4个方面开展该解毒酶在适应不同宿主植物中的功能与表达调控的分子机理研究:1) 斜纹夜蛾SlGSTE1对不同宿主植物次生物质应答与降解作用; 2) SlGSTE1功能的RNA干扰分析;3) 诱导SlGSTE1表达的信号途径和调控机理;4) SlGSTE1基因表达的转录后水平调控机理。通过研究斜纹夜蛾中 SlGSTE1的功能、解毒途径和表达调控机理,为探索以SlGSTE1为靶标的害虫防治方法和策略、降低抗药性和减少环境污染,提供理论依据和技术支撑。

英文摘要: Insect generalists can adapte a wide of range of plants by their complicated detoxifying-enzyme systems, in which different enzyme isoforms detoxify different plant allelochemicals. This mechanism also results in the different sensitivity of the insects to insecticides. This causes difficulties to the development of the strategies of pest management in which a single detoxifying enzyme is used. There is few report on the study of the regulatory pathways of the detoxification enzymes of insects feeding on different host plants and no study on the transcriptional and post-transcriptional regulation of the expression of these enzymes. In this project, we will focus on a detoxification enzyme, glutathione S-transferase SlGSTE1 in Spodoptera litura and study the function and molecular machanism of expression regulation of this gene. The study will include four aspects: (1)The indcued expressions and detoxicification function of SlGSTe1 in response to different plant allelochemicals; (2) the function analysis by RNAi; (3) signal pathway analysis of SlGSTe1 expression in response to different plant allelochemicals; (4) the mechanism of post-transcriptional regulation of SlGSTe1.
