中文摘要: 水貂作为一种珍贵的毛皮动物,其被毛颜色是影响其裘皮品质的重要质量性状。TYRP1基因作为影响动物被毛颜色变异的重要候选基因,近年来得到广泛重视。本项目拟在前期对山羊和狐狸TYRP1基因研究成果基础上,开展水貂TYRP1基因研究。研究内容包括:(1)利用比较基因组学等手段研究不同毛色水貂TYRP1基因的变异类型及其与毛色的相关性;(2)开展不同毛色水貂皮肤组织TYRP1基因mRNA及其编码蛋白差异表达研究;(3)利用双荧光素酶报告系统等技术研究TYRP1基因启动子区变异及其对基因表达影响;(4)利用EMSA、ChIP等技术研究TYRP1基因的转录因子及其对基因表达影响;(5)开展TYRP1功能获得/缺失研究探讨其调控黑色素形成机制。研究结果不仅可以为揭示TYRP1基因调控水貂毛色分子遗传机制提供科学理论依据,同时对于揭示水貂毛色的遗传多样性、提高水貂分子育种水平具有重要的理论和实践意义。

英文摘要: The coat color of mink, a kind of precious fur animal, is important qualitative character for affecting mink pelt quality. TYRP1 gene is an important candidate gene affecting animal coat color variation and has attracted widespread attention in recent years. Based on the previous work of goat and fox TYRP1 gene, this project will carry out the study of mink TYRP1 gene. Firstly, we will investigate the variation of TYRP1 and its correlationship with coat color by using comparative genomic etc. Then, we will carry out the skin differentiation expression of TYRP1 mRNA and its coded protein. By using the dual luciferase reporter assay system etc, TYRP1 gene promoter region variation and its impact on gene expression will be studied. Additionally, we will investigate the TYRP1 gene transcription factors and their impact on gene expression by using EMSA, ChIP, etc. To understand the regulation mechanism of TYRP1 for the formation of melanin, we will evaluate effect of overexpression and knockdown TYRP1 in melanin cells. Not only will the results provide the scientific base for regulation of TYRP1 gene on molecular genetic mechanism of mink coat color, but also they have important theoretic and practical sence for study the genetic diversity of mink coat colour, and for improving the molecular breeding of mink.
