中文摘要: 肿瘤干细胞具有自我更新和分化为各级肿瘤细胞的能力。研究显示一些肿瘤细胞能在干细胞和非干细胞状态之间转换,但对肿瘤细胞塑型的分子机制仍知之甚少。我们的预实验结果显示G3BP2能促进NSCLC细胞出现干细胞样特征,提示G3BP2可能参与调控NSCLC塑型。同时G3BP2可能与SART3结合,而文献报道后者在胚胎干细胞中调控核心转录因子,维持干细胞特性。据此我们提出假说:G3BP2与SART3形成复合物,作为RNA结合蛋白与OCT4、Sox2和Nanog mRNA相结合并调控其表达,从而参与NSCLC塑型。我们将采用多种NSCLC细胞株,采用免疫组化、PCR、western blot、Co-IP、RIP等技术从分子、细胞、组织和动物水平验证上述假设。本研究能进一步完善肿瘤干细胞对肿瘤发生发展的调控机制,并为临床干细胞治疗和逆转耐药提供新的靶点。

英文摘要: Tumors contain cancer stem cells (CSCs) that are thought to be capable of replenishing both themselves and populations of non-CSCs. Recent evidence suggests that some cancer cells can switch between a non-CSC and CSC-like state, the molecular mechanisms underlying this plasticity remain poorly understood. Our preliminary data have shown that G3BP2 is essential for the maintenance of a sub-population of NSCLC cells with CSC-like properties. Furthermore, G3BP2 has physically interaction with SART3, which is essential in regulating the core transcription factors in hESCs. Take together, we hypothesize that G3BP2 interacts physically with SART3 and combines with OCT4, Sox2和Nanog mRNA, which regulates NSCLC plasticity. We will verify the hypothesis by using IHC, PCR, western blot, Co-IP, RIP et.al. Our study will support the potential role of G3BP2 as a “master switch” controlling NSCLC plasticity.
