中文摘要: 淫羊藿黄酮(Epimedium-derived flavonoids, EFs)有防治绝经后骨质疏松的作用。淫羊藿苷(Icariin)是EFs最主要的生物活性成分。体外细胞实验表明Icariin同时具有促进成骨和抑制骨吸收的双重作用,可以有效的治疗骨质疏松,但这种双重作用的机制尚不明确。我们前期工作捕捉到和推测OPG和Wnt/β-catenin信号通路可能参与这种双重调节作用,但具体机制并不完整和明晰。因此,本课题研究项目将在前期工作的基础上,通过体外和体内两方面深入和全面的研究,寻找、明确和归纳Wnt/β-catenin信号通路和OPG在Icariin双重调控骨代谢中的信号和分子机制,包括Wnt/β-catenin信号通路和OPG在研究中可能呈现的基于细胞信号网络化调控基础上的相互调节机制。为Icariin作为中药提取小分子治疗骨质疏松提供坚实的实验证据和理论基础。

英文摘要: Epimedium-derived flavonoids(EFs) have been play a role in the prevention of Post- menopause Osteoporosis, and the bioactive effect of this matter is mainly achieved by the key component called Icariin in EFs.In vitro study, it indicates that icariin could promote bone formation and inhibit bone resorption.However, the mechanism of the dual effects ramains elusive. Our previous studies found that OPG and Wnt/β-catenin signal pathway maybe been involved in the dual effects of Icariin. On the base of our preliminary studies, through the studies in vitro and in vivo,the further studies will try to find and clarify the molecular and signal pathway mechanism of OPG and Wnt/β-catenin signal pathway when they work in icariin control bone metabolism dually. Specially, the cross-talking between OPG and Wnt/β-catenin signal pathway which base on the network control of Cell signal would been understood. At last, a new micromolecule drug of icariin will be developed for treatment of Osteoporosis based on the mechanism aforementioned .
