中文摘要: 生物细胞力学性质的测量是微尺度下细胞研究及微流控芯片发展的基础之一。本项目以微流控芯片技术为研究平台,通过对非线性超声辐射场内细胞的包壳模型的受力及其运动规律的基础科学研究,发展用于测量细胞局部力学性质的声辐射测量理论。具体内容包括:建立细胞的包壳结构所受声辐射力的理论模型;分析局部各结构层弹性模量与细胞整体弹性模量间的关系,发展超声辐射测量细胞局部弹性模量的测量原理;构建相应的超声微流控芯片系统,实现对典型生物细胞弹性模量的实验研究。本项目旨在发展微尺度下的声辐射测量理论,实现对细胞局部力学性质的研究。其研究结果可将现有的对细胞整体的超声辐射研究手段拓展到对细胞局部力学性质的研究上;其所测量的细胞局部力学特性,可充实细胞力学数据库,有助于深入分析各类致病细胞的力学特性与其致病机理间的关系;同时拓展微流控芯片中超声分离、提纯、与检定技术提供理论与技术基础。

英文摘要: The measurement of the mechanical properties of biological cells is one of the foundations of the cell researches in micro scale and it also serves as the fundamental characters of the development of the micro-total analysis system. In this project, we investigate the forces acting on the cells and the consequent motion characters of such cells suspended within the microfluidic system where the nonlinear ultrasonic radiation field stands; furthermore, we develop the measurement method of the cell's mechanical modulus using the ultrasound radiation field within the microfluidic system. The detailed content is arranged as follows: To develop the theoretical model which can describe the acoustic radiation force acting on the cell that can be simplified as a shelled spherical particle; To compare the shell spherical model and the uniform compressible spherical model, and establish the relationship between the cell's global elastic modulus and the cell's local elastic modulus of each structural layer; To develop the ultrasonic measurement method that can measure the cell's elastic modulus(global and local); To design and fabricate the corresponding microfluidic system and realize the measurement based on our proposed method.
