中文摘要: 细胞核移植是实现体细胞克隆的主要手段,如何通过微操作方法的改进,提高核移植成功率是基于微操作机器人系统的克隆实验面临的挑战。从微操作角度,影响核移植成功率主要有三个因素:卵细胞发育状况、核移植操作持续时间及创伤程度。这三个因素对微操作的挑战分别是:细胞弹性在线测量,确定性操作结果的细胞拨动方法和从可行的刺破细胞透明带方案中寻求最优的操作方法。进一步研究可以发现三个挑战的弹性本质:细胞发育程度检测,是一个弹性在线测量问题;细胞拨动结果不确定,是细胞弹性形变造成的;刺破细胞透明带是弹性形变极限问题。本项目定位在基于细胞弹性的微操作方法研究。对应提高核移植成功率的三个挑战,研究内容分别为:1)基于微操作的细胞弹性模量测量方法研究; 2)基于弹性的细胞拨动方法与最佳拨动策略研究;3)刺破细胞透明带过程建模与最优控制; 4)面向核移植成功率的家畜克隆实验研究。

英文摘要: As the nuclear transplantation is the crucial approach to realize somatic cell cloning, the facing challenge to the cloning experiment based on the micro manipulator robot system is how to improve the micromanipulation method in order to increase the nuclear transplantation success rate. From the view of micromanipulation, three main factors, which include the oocyte development status, the duration of nuclear transplantation operation and its corresponding wound degree, may exert impact on the nuclear transplantation success rate. These three factors have raised challenges to the micromanipulation, that is, the cell elastic modulus on-line measurement, the way of stirring the cell which would generate deterministic operation result and to seek for the optimal operation method from the feasible schemes of piercing the oocyte zona pellucida. The further study will reveal the essence of the above three challenges:the detection of cell development degree is a question of its elasticity on-line measurement; the uncertainty of stirring the cell is attributed to its elastic deformation; piercing the oocyte zona pellucida is a question of its limit elastic deformation.This project aims to study the operation method based on cell elasticity.The three research contents corresponding to the challenges of improving the nuc
