中文摘要: HtrA蛋白在微生物抵抗胁迫环境中扮演着重要的角色,然而,其如何调控并参与微生物适应胁迫环境的分子机理目前尚缺乏系统深入的认识。本研究利用前期蛋白质组研究中发现的与丁醇耐受性相关的靶蛋白-HtrA,在模式梭菌Clostridium acetobutylicum ATCC 824中进行htrA基因的过表达和敲除研究,揭示其与丁醇耐受性的关系,并利用蛋白质组学技术、real-time PCR和生理学分析检测突变菌株和靶蛋白响应丁醇浓度时蛋白质组、RNA水平的差异表达以及生理学变化,推测HtrA蛋白调控丁醇耐受性的分子机制;此外,在大肠杆菌中过表达HtrA蛋白,检测菌株丁醇耐受性的变化,进一步验证其生理学功能。研究结果将初步阐明HtrA蛋白介导的丁醇耐受性分子机制,为C. acetobutylicum 菌种改造以及新型工艺的开发提供理论基础,同时为其它醇类生产菌株的改造提供理论指导和借鉴。

英文摘要: It has been previously reported that HtrA protein plays an important role in tolerance to environmental stress of microorganism. Currently, the mechanism of HtrA protein participation and adaptation to environmental stress in microorganism lacks system research. The project used butanol tolerance related protein-HtrA in previous research. Through overexpression and konckout of htrA gene in Clostridium acetobutylicum ATCC 824, it revealed the relationship between htrA and butanol tolerance. We used proteomics technologies, real-time PCR and physiological analysis to detect their response of all strains and target protein HtrA to butanol stress. It can speculate that the molecular mechanism of HtrA protein regulation of butanol tolerance. In addition, overexpression of htrA gene in Escherichia coli and analysis butanol tolerance of mutant strain can be evaluated its physiological function. These results allow us to obtain the preliminary elucidating of the molecular mechanism of HtrA protein mediation butanol tolerance. It will aid the strain improvement of C. acetobutylicum and development of the new process, meanwhile, provide theoretical support for improvement of alcohols- producing strains.
