中文摘要: 芝麻是我国传统特色油料作物,芝麻素(sesamin)是芝麻中特有的功能性成分,具有提高芝麻油品质和调节人体多种生理功能作用。芝麻素的研究早已成为国内外研究热点。挖掘高芝麻素基因资源、开发有效功能标记用于高品质芝麻育种辅助选择已是芝麻育种改良和产品深加工等产业发展的迫切需要。本研究基于已发掘的芝麻素含量差异群体,利用同源克隆获取该群体芝麻素合成基因CYP81Q1全长序列;通过半定量RT-PCR研究芝麻素合成基因表达模式和调控机理。在此基础上,采用基因-表型关联分析方法获得芝麻素含量关联标记,进而利用芝麻素杂交分离群体验证并获取高芝麻素功能标记,以期形成芝麻素分子辅助育种体系。本研究将对芝麻重要经济潜能基因发掘、分子辅助育种等提供技术和关键材料支撑,具有重要理论和实际意义。

英文摘要: Sesame is a conventional and special oil crops in China. As a special functional component in sesame, sesamin is vital to enhance sesame oil quality and adjust human physiological functionment. Sesamin research have became the hot research area in China and abroad. Identification of sesame resources with high sesamin content, developing and appling effective functional markers to molecular assistant selection in sesame quality breeding, have been exigent necessary in sesame improvement and sesame industry development. Basing on the population varied in sesamin content, this study aims to obtain the full sequence of sesamin synthase gene CYP81Q1 by homologous cloning, and investigate the gene expressing and regulation pattern by semi-quantitative RT-PCR. Furthermore, this study will get functional markers of sesamin content using gene-phenotype association analysis, then verify the markers by sesamin segregating population (F2), in expectation of establishing sesamin molecular breeding system. Therefore, the study will provide technique and key material support to discover the important economic potential genes and sesame breeding methodology innovation such as marker assisted selection.
