中文摘要: 云南软米是云南特有的一种优质米资源,其独特蒸煮和食味品质深受各族人民喜爱,具有广阔市场前景,是稻米品质改良的优异遗传资源。本项目以前期收集和保存的47份云南软米资源为材料,拟通过测定表观直链淀粉含量、胶稠度、糊化温度及淀粉粘滞度(RVA谱值),系统评价云南软米资源淀粉品质特征;同时基于水稻直链淀粉合成酶(GBSS),可溶性淀粉合成酶(SSS),淀粉分支酶(SBE)以及淀粉去分支酶(DBE)已报道的16个基因序列,采用单链构象多态性分析(SSCP)及测序技术,开发用于云南软米资源遗传研究的特异标记;利用本研究开发的标记及已有淀粉合成基因标记对云南软米资源进行基因型鉴定,并结合表型数据对淀粉品质性状进行关联分析,研究淀粉合成关键酶基因位点变异对淀粉理化特性的影响。为揭示云南软米资源淀粉品质形成的遗传机制及分子标记辅助选择提供理论基础。

英文摘要: Yunnan soft rice are the favorable genetic resources for quality improvement, which are popular with local residents by unique cooking and eating quality. In this research, 47 Yunnan soft rice varieties from the collection and conservation in gene bank will be analyzed. By detecting the apparent amylose content, gel consistency, gelatinization temperature and starch viscosity(RVA profile value), starch and eating quality in Yunnan soft rice resources will be evaluated. Meanwhile, based on the 16 gene sequences of rice amylase synthase(GBSS), soluble starch synthase(SSS), starch branching enzyme(SBE) and starch debranching enzyme(DBE), markers will be developed by SSCP and sequencing technique for polymorphism analysis in Yunnan soft rice resources. Then, these markers above and other markers reported before will be used for genotyping analysis in Yunnan soft rice resources. Finally, elite alleles associated starch quality will be discovered by association analysis between phenotype and marker locus. These results will be helpful for understanding the genetic mechanism of starch quality formation in Yunnan soft rice and molecular maker-assisted selection.
