中文摘要: 家系设计是复杂疾病关联分析中重要的研究设计方式之一。与群体病例对照设计相比,家系设计的优势在于能够有效控制群体分层等混杂因素所导致的统计分析的偏差,但该设计下统计检验的功效通常偏低,所以提高检验功效是家系关联分析中的一个关键问题。在基因模型已知的条件下,传递不平衡类型检验(即TDT-type检验)具有最大的检验功效,但在基因模型错误指定时,该类型检验通常有较大的功效损失。本项目研究定量家系关联分析中利用偏离哈代-温伯格平衡的信息来提高检验功效,并构造对基因模型指定稳健的有效检验。当性状数据服从正态分布时,针对核心家系和其它家系设计提出并研究三类基于TDT-type检验的稳健有效检验;当性状分布未知时,研究对基因模型和性状分布双重稳健有效的非参数检验方法。关于定量性状家系数据的稳健有效方法研究,将为基于家系的全基因组关联分析提供有效的统计方法,具有较为广泛的实际应用价值。

英文摘要: Family-based design is one of the commonly used approaches in genetic association studies. Compared with population-based case-control design, family-based designs can effectively control the bias caused by confoundings such as population stratification, however, power of such designs is usually low comparing with case control designs with the same sample size. Therefore investigation of powerful statistical methods is a key issue in family-based association studies. If the genetic model or mode of inheritance is known, then TDT-type tests(Transmission Disequilibrium Tests)are most powerful in detecting association. However, if the genetic model is misspecified, TDT-type may substantially lose power, i.e., TDT-type tests lack robustness property to the genetic model specification. This study aims at constructing robust-efficient tests for quantitative data analysis in family designs by using the deviation from the Hardy-Weinberg equilibrium. When the trait data is normally distributed and the design is nuclear family or other family-based design, we propose three robust-efficient tests constructed from TDT-type tests and study their properties.
