中文摘要: 家蚕细小病毒样病毒(Bombyx mori parvo-like virus, BmPLV)是目前已知的唯一一例自编码DNA聚合酶的单链线性DNA病毒,也是首例动物二分病毒,其基因表达调控及复制机理未明。我们的前期研究表明BmPLV-Z NS1 是一个多功能蛋白,具有ATPase活性、特定DNA序列结合以及helicase活性,预示该蛋白与病毒DNA复制密切相关。但NS1蛋白的表达调控模式和其相互作用的靶分子尚不清楚。本项目拟开展(1)ns1基因转录本及其启动子分析;(2) NS1蛋白转录激活功能分析;(3)分析NS1蛋白磷酸化及磷酸化与功能的关系;(4) NS1蛋白的互作蛋白的分离鉴定。作为与DNA复制密切相关的多功能蛋白,揭示NS1蛋白的表达调控特征及其在宿主细胞内相互作用的靶分子,对阐明BmPLV病毒的基因表达调控及复制机制具有重要意义。

英文摘要: To the best of our knowledge, Bombyx mori parvo-like virus (BmPLV) is the unique virus which encodes a DNA polymerase and posses two linear single-strand DNAs, but the mechanisms of the virus gene expression and virus replication are unknown. Our previous research showed that the non-structrue protein NS1 combined multiple functions such as ATPase activity, helicase activity and site-specific binding to a DNA recognition motif, indicting that it plays a key role in the replication of virus DNA. However, the mode of the expressing regulation and the interacted target proteins of NS1 are unknown. In this project, following issues will be resolved in the future: (1) to analyze the promoter activity and translational mode of ns1 gene, (2) investigating the activity of promoter trans regulation of NS1, (3) determining the phosphorylation site of NS1 and the association between the phosphorylation and its function, (4) characterization of the target proteins interacting with NS1. As a key multifunctional protein closely involved in virus DNA replication, we expect that elucidating the mode of NS1 expressing regulation and the target proteins interacting with NS1 will help us to understand the mechanism of the gene expressing regulation and replication of virus.
