中文摘要: Toll样受体(TLR)是在人、小鼠上最新发现的与宿主免疫识别有关的分子,能广泛地、特异性地识别病原或其产物,而激活宿主的先天性和获得性免疫应答。本项目以发现和揭示新的免疫识别机制为出发点,选择能基本代表Toll样受体家族重要功能的分子―#8213;猪TLR7为研究对象,利用分子生物学、差异表达分析和生物信息学技术,研究其分子结构、组织细胞分布和差异表达特征;建立其模式识别细胞模型体外评价系统,选择口蹄疫病毒、蓝耳病病毒和CpG DNA等代表性的病原模式分子,通过抗体阻断、sRNA干扰以及实时定量PCR技术等手段,研究其免疫识别病原相关模式分子的种类、结构特点以及诱发的免疫学反应现象,探讨猪TLR7对ssRNA病原相关分子模式识别的种属特异性和差异性,初步揭示ssRNA病毒普遍入侵宿主引起感染或免疫应答的分子基础,为猪疫病发生和控制技术研究提供理论依据。

英文摘要: Toll like receptors (TLRs) are molecules associated with host immune recognition, which have been discovered recently in human and mouse. These molecules are able to recognize various types of particular pathogen or its products, and further activate the host innate and adapted immune response. The aim of this research is to identify and understand the mechanism underlying the porcine TLR7 (pTLR7), the representative molecular of TLR family, mediated immune recognition. In this proposed study, we will evaluate the molecule structure, tissue distribution and differential expression condition of pTLR7, by using several techniques related to molecule biology, differential expression analysis and bioinformatics analysis. To establish in vitro cell based pattern-recognition evaluation model system, the typical pathogen associated molecule patterns, including foot-and-mouth disease virus (FMDV), porcine reproductive and respiratory syndrome virus (PRRSV) and CpG DNA will be chosen to study the type and structure of pathogen associated molecule patterns, as well as induced immune response through antibody blocking, sRNA interference and real time PCR. These studies will define the species specificity and discrepancy of pTLR7-mediated recognition of single-stranded RNA (ssRNA), will elucidate the general molecular basis of the infection and immune response caused by ssRNA virus, and will also provide new information for the epidemiology study and prevention of swine diseases.
