中文摘要: 液泡氢ATP酶(V-ATPase)是一个多亚基组成的复合体酶,主要通过ATP驱动的质子泵活性调控细胞内pH平衡,参与许多重大的生理活动。B亚基不仅是ATP水解的重要功能位点,并且可以结合微丝骨架,在调控V-ATPase活性中起着至关重要的作用。但对于植物V-ATPse B家族成员的研究非常匮乏。本研究拟利用分子生物学、细胞生物学、生物化学、遗传学等方法,以植物模式材料――拟南芥为研究材料,首次系统的研究植物V-ATPase B亚基家族的3个不同成员各自的基因定位、生理生化功能、作用分子机制等内容,旨在揭示植物V-ATPase B亚基如何参与调控植物不同生理活动及其分子机制。本项目研究成果不仅有利于揭示V-ATPase作用分子机制,并且还有可能为揭示植物细胞极性生长机制、植物信号传导途径以及抵御非生物胁迫等相关研究提供新知识、新材料和新方法,具有非常重要的理论研究意义和实际应用价值。

英文摘要: Vacuolar H+-ATPase is an enzyme complex composed of many subunits to take part in many crucial physiological events via regulating intracelullar pH homeostasis by the activities of proton pump drived by ATP. The B subunit is not only the pivotal function site of ATP hydrolysis,but also can bind to actin cytoskeleton, so it plays an inportant role in V-ATPase activity regulation. However, there are few reports about the members of V-ATPase B subunit in plant.For this reason, it is the first time to intend to use model plant Arabidopsis systematically to focus on the genes localization, physiological and biochemical functions as well as molecular mechanisms of three distinct members of V-ATPase B subunit via experimental approaches of molecular biology,cell biology,biochemistry and genetics. Our final research goal is to illustrate the molecullar mechanisms how V-ATPase B subunit to regulate plant physiological activities. This program results might also supply the new theory, meterial and method for related research on cell polar growth, signal transduction pathway and abiotic stress defense in plant, therefore, the project is provided with extremely important theoretical research significance and application value.
