中文摘要: 动物营养学与繁殖学交叉学科研究正在兴起形成新的研究领域。申请者团队前期研究已证实维生素E对公羊繁殖功能有着明显的促进作用,但其分子调控机制尚未完全明确。在前期研究基础上,本研究拟构建绵羊睾丸原代细胞,经α-生育酚处理后,综合利用目前国际最新组学技术,筛选绵羊睾丸细胞中同维生素E相关的microRNA、mRNA及蛋白因子;然后通过生物信息学对相应差异表达因子的功能和Pathway网络进行分析,建立与维生素E相关的microRNA-mRNA-蛋白质分子调控网络。随后设计不同水平维生素E日粮饲喂公羔,利用荧光定量PCR和Western-blot技术从microRNA、mRNA和蛋白质三个层次在机体水平检测相应分子调控网络中重要因子的表达特点,从而确定维生素E调控睾丸功能的关键信号通路。本研究对揭示维生素E调控睾丸功能的分子机理及建立提高繁殖性能的营养调控技术有着重要的科学意义和理论价值。

英文摘要: Cross disciplinary research about animal nutrition and reproduction is becoming a new research area. It has been proved by our previous research that vitamin E played an important role in regulating the reproduction function of ram, however, the molecular mechanism is still poorly understood. Therefore, the primary ovine testis cells will be first constructed and after treated with α-tocopherol, differentially expressed microRNA, mRNA and proteins will be identified using the latest omic technology. Then the functions, signal networks, and regulatory networks of these factors will be analyzed by using bioinformatic analysis. Important factors of these networks will be selected for verifying their functions at microRNA, mRNA and protein levels by using techniques such as real-time PCR and Western blot. The results of this study will uncover the molecular mechanisms of vitamin E on ovine testis function, as well as provide guidance for establishing nutritional regulation technology on reproduction performance in sheep.
