中文摘要: 灰霉病是危害辣椒等蔬菜作物生产最严重的病害之一,WRKY转录因子在植物免疫进程中具有重要调控作用,然而目前WRKY转录因子在辣椒灰霉抗性应答中的调控机理还不清楚。申请人前期研究发现辣椒CaWRKY转录因子在抗病响应中发挥重要调控作用,并鉴定发现CaWRKY基因在辣椒灰霉抗/感材料中转录水平存在差异,进而筛选获得6个潜在参与辣椒灰霉应答调控的CaWRKY成员。基于此,提出CaWRKY参与辣椒灰霉病抗性转录调控的假设。本项目在分析辣椒全基因组61个CaWRKY蛋白/基因结构、系统进化等基本属性基础上,利用qRT-PCR、VIGS及转基因等技术研究CaWRKY在辣椒灰霉病抗性中的功能,并结合酵母单/双杂交技术分析CaWRKY与其它灰霉应答信号路径元件的相互作用,定位CaWRKY在调控辣椒灰霉胁迫响应中的关键信号节点。总之,本项目的实施将为解析CaWRKY参与调控辣椒灰霉病抗性机理奠定基础。

英文摘要: Gray mold caused by Botrytis cinerea is one of the most severe diseases in the production of pepper and other vegetable crops, WRKY transcription factors play important role in plant immunity system. However, the molecular mechanism of WRKY transcription factors in response to B. cinerea is still unclear. Our previous study demonstrated that pepper CaWRKYs may be involved in the response of desease resistance, and the transcript levels of CaWRKY genes were significantly different between B. cinerea resistant/susceptible lines, 6 candidate CaWRKYs potentially involved in B. cinerea response were identified. Thus, We proposed the hypothesis that CaWRKYs play critical roles in the transcriptional regulation of B. cinerea responding process in Pepper. Based on the characteristic analysis of 61 CaWRKYs, using technical means including qRT-PCR, VIGS and transgene, functional analysis of CaWRKYs were conducted. Furthermore, through yeast one/two-hybrid system, the interactions between CaWRKY and other components of B. cinerea response pathway were studied. In short, the implementation of this project would provide theoretical basis for the B. cinerea resisting regulation mechanism modulated by CaWRKY transcription factors in Pepper.
