中文摘要: 小黑豆是我国独特的抗线虫资源,在国际抗大豆胞囊线虫育种研究领域受到了广泛的关注,美国仅用几个来自中国的小黑豆品种就育成了100多个抗线虫品种。我国具有近百个抗线虫的小黑豆品种,申请者在前期抗线虫基因归类和遗传分析,表达谱分析的基础上,拟在大豆胞囊线虫3号生理小种的选择压力下选择位于C2染色体上的大豆谷胱甘肽过氧化物酶基因GmGpx1和位于A2染色体上的GmHs1Pro-1基因对不同的小黑豆品种在表达水平上进行功能分析,分析它们在大豆胞囊线虫侵染条件下的表达模式;利用RNAi和基因过表达等技术,深入研究目的抗性基因的作用;阐明这些基因在胞囊线虫抗性免疫防御中的作用及调控机制。该项研究将为小黑豆抗大豆胞囊线虫基因资源发掘提供理论基础,筛选存在于小黑豆品种中可用于基因工程育种开发的靶标基因,为有效利用我国小黑豆资源培育抗大豆胞囊线虫品系提供新的选择性标记,具有重要的理论意义和实践意义。

英文摘要: Small black soyabean is the unique nematode resistant resources of China.The black coat soybeans had been widespread concerned in the international resistant breeding of soybean to Heterodera glycines(Soybean Cyst Nmeatode,SCN). Only a few small black soybean varieties were used to breed more than 100 resistant varieties of resistant to SCN in United States. There are nearly 100 varieties of small black soybean in our country. For the basis of research for small black soybean against to SCN, the soybean glutathione peroxidase gene GmGpx1 on chromosome C2 and the soybean nematode resistance gene GmHs1Pro-1 chromosome A2 were studied under the selection pressure of the SCN Race 3 ( HG 5,7 )in different small black soybean varieties in the expression level of functional analysis, analysis of their expression pattern in the soybean cyst nematode infection condition. Acording the use of RNAi and gene technology, the in-depth study will be done in these genes in the cyst nematode resistant function; further study of resistance related signaling pathways against nematode gene regulation, the elucidation of these genes in the cyst nematode resistance in immune defense function and regulation mechanism. The study will provide small black soybean resistance to soybean cyst nematode gene resource discovery provides the theoretical basis, to screen for the presence of small black soybean varieties can be used in gene engineering breeding and development of the target gene, effective use of small black soybean breeding for resistance to soybean cyst nematode strains provide new selective markers. It has the important theory and practical significance for the researches.
