中文摘要: Nakamura 等人在水稻基因组中预测到6个潜在的光敏色素相互作用因子基因OsPIFs。根据它们所编码蛋白质中DNA结合域氨基酸的相似性,可将其分为3亚类,即OsPIL11/PIL12、OsPIL13/PIL14和OsPIL15/PIl16。但是,这些OsPIFs因子如何参与光敏色素介导的光形态建成尚不清楚。 本项目以OsPIL11、OsPIL13和OsPIL15为代表,从以下几个方面系统分析它们在水稻光形态建成中的作用:(1)鉴定3个OsPIFs的亚细胞定位及其与水稻光敏色素之间物理学上的相互作用;(2)分析光敏色素介导的红光或远红光对3个OsPIFs基因表达的影响;(3)分析3个OsPIFs基因过量表达和RNAi转基因水稻的光形态建成特征,阐明不同OsPIFs在水稻生长发育中的作用。本项目研究结果将为全面揭示水稻光信号传导网络提供突破口,同时为改良水稻相关农艺性状的分子设计奠定基础。

英文摘要: Nakamura et al. identified six putative genes encoding phytochrome-interacting factors (designated OsPIL11-OsPIL16) via an extensive inspection of the entire genome sequence databases of rice (Oryza sativa). Based on their similarity of amino acid sequence of DNA-binding domain, six OsPIFs were classified as three subclasses of OsPIL11/12, OsPIL13/14 and OsPIL15/16. However, how OsPIFs act in phytochrome-regulated photomorphogenesis remains elusive. In this project, we choose OsPIL11, OsPIL13 and OsPIL15 as candidates and analyze their roles in phytochrome-mediated photomorphogenesis in rice through the followed aspects. (1) To examine the subcellular localization of OsPIL11, OsPIL13 and OsPIL15 as well as the interaction between OsPIFs and rice phytochromes. (2) To analyze the effects of phytochrome-mediated red or far-red light on OsPIL11, OsPIL13 and OsPIL15 expression at both transcriptional levels and translational levels. (3) To uncover the roles of OsPIL11,OsPIL13 or OsPIL15 in rice growth and development by characterizing the photomorphogenesis of transgenic rice plants with either overexpressed individual OsPIFs or reduced OsPIFs.
